چکیده

We conducted a case-control study to investigate the role of ABCB1 C3435T (rs1045642) and C1236T (rs1128503) gene polymorphisms in the susceptibility to acute myeloid leukemia. The polymerase chain reaction (PCR) coupled with restriction fragment length polymorphism (RFLP) method was carried out to genotyping ABCB1 C3435T and C1236T gene polymorphism. Statistical analysis was conducted using the SPSS 17.0 package (SPSS Inc., Chicago, IL, USA). Using Chi-square test, a statistically significant difference was found between acute myeloid leukemia patients and control subjects with respect to ABCB1 C3435T (χ2=8.04, P=0.02). By unconditional logistic regression analysis, we found that the TT genotype of ABCB1 C3435T was associated with an increased risk of acute myeloid leukemia compared to the CC genotype (OR=2.24, 95% CI=1.22-4.12, P=0.01). In dominant model, the CT+TT genotype of ABCB1 C3435T was correlated with a higher risk of acute myeloid leukemia than the CC genotype (OR=1.68, 95% CI=1.07-2.65, P=0.02). In the recessive model, the TT genotype of ABCB1 C3435T contributed to the development of acute myeloid leukemia when compared to CC+CT genotype (OR=1.74, 95% CI=1.04-2.95). In our study, we suggested that the ABCB1 C3435T genetic polymorphism could influence the risk of developing adult acute myeloid leukemia in co-dominant, dominant and recessive models.

Role of ABCB1 C1236T, G2677T, and C3435T genetic polymorphisms in the development of acute leukemia in a Chinese population.

We carried out a case-control study to examine the relationship between the ATP-binding cassette subfamily B member 1 (ABCB1) gene polymorphisms C1236T, G2677T, and C3435T and risk of acute leukemia in a Chinese population. Between May 2013 and April 2015, we recruited 164 acute leukemia patients and 285 healthy controls, and determined polymorphism genotypes by polymerase chain reaction-restri...

Association of ABCB1 polymorphisms and ulcerative colitis susceptibility.

OBJECTIVE Multi-drug resistance gene 1 (ABCB1) is closely related to bowel diseases. Therefore, our study was aimed to evaluate the correlation between ABCB1 polymorphisms (C1236T and C3435T) and ulcerative colitis (UC) susceptibility. METHODS A total of 61 UC patients and 64 healthy people participated in the study. Genotyping was conducted with the method of polymerase chain reaction-restri...

Impact of ABCB1 Gene Polymorphisms and Smoking on the Susceptibility Risk of Chronic Myeloid Leukemia and Cytogenetic Response

Background: Imatinib mesylate (IM), a strong and selective tyrosine kinase inhibitor, has been approved as the front line of treatment in chronic myeloid leukemia (CML) patients. In spite of satisfactory results of imatinib in the treatment of patients with CML, patients with treatment failure or suboptimal response developed resistance that might be because of pharmacogenetic variants. This st...

Association of Acute Lymphoblastic Leukemia and MDR1 Gene Polymorphism in an Ethnic Iranian Population

Background: The frequency of the multi-drug resistance 1 (MDR1) gene C3435T polymorphism differs in various ethnical populations such as Asian, African, and Caucasians populations. A silent C3435T polymorphism in exon 26 of MDR1 has been reported to be associated with a decreased expression of P-gp in TT genotypes carriers compared with CC genotypes carriers. Materials and Methods: To evaluate ...

Common ABCB1 polymorphisms associated with susceptibility to infantile spasms in the Chinese Han population.

Infantile spasms are a severe epileptic encephalopathy with a variety of etiologies that occur in infancy and early childhood. Subjects with infantile spasms are at a higher risk for evolving into intractable epileptic spasms, tending to be refractory to conventional antiepileptic drugs. Genetic polymorphisms of the P-glycoprotein-encoding gene ABCB1 are suspected to be associated with pharmaco...
